Dimebon (latrepirdine) is an investigational compound currently in Phase 3 development for the treatment of Huntington disease and Alzheimer's disease.
The mechanism of dimebon (latrepirdine) is distinct from other Huntington disease and Alzheimer's disease agents. In preclinical studies, dimebon has been shown to enhance neuronal survival in the presence of cellular stress, potentially by stabilizing and improving mitochondrial function.

Huntington disease trial
The DIMOND study, a multicenter, randomized, double-blind, placebo-controlled Phase 2 study of dimebon (latrepirdine), was recently completed. The DIMOND study tested the safety and tolerability of dimebon treatment and the effect of dimebon on cognitive, motor, and overall function in patients with Huntington disease. Based on the results of this study, the Horizon Trial was initiated.
Alzheimer's disease trials
In the first pivotal clinical trial of dimebon (latrepirdine) in patients with mild to moderate Alzheimer's disease, the following points were examined:
- Cognition and memory
- Self-care and daily function
- Behavior
Based on the results of this recent study, the Connection Study was initiated. The Connection Study is a global Phase 3, double-blind, placebo-controlled trial examining the efficacy and safety of dimebon (latrepirdine) in a large cohort of patients with Alzheimer's disease. A second Phase 3 trial, the Concert Study, is currently enrolling. The Concert Study is a clinical research study of 1050 patients to test the effects of dimebon (latrepirdine) in patients with Alzheimer's disease who are currently taking Aricept® (donepezil HCI tablets). The purpose of the Concert Study is to determine whether dimebon safely improves cognition (thinking and awareness), memory, daily functioning, behavior, and the ability to care for oneself.
